Comparative study on the effects of substrate stiffness on cell morphology and focal adhesion expression between hMSCs and AFSCs

چکیده
Results Staining and imaging showed numerous clear focal adhesions had been formed for both cell types and the general morphology of the cells represented that of healthy cells with a flatly spread appearance when cultured on the glass cover-slips. On the collagen films, the cells showed poor focal adhesion formation and very diffuse staining, but no difference was observed as a function of crosslinking technique. Furthermore, the low stiffness also allowed the cells to contract giving them a narrow, elongated morphology.

The Effects of Iron Oxide Nanoparticle on Differentiation of Human Mesenchymal Stem Cells to Osteoblast
Introduction: IIron oxide nanoparticles (IO NP) have an increasing number of biomedical applications. To date, the potential cytotoxicity of these particles remains an issue of debate. Little is known about the cellular interaction or toxic effects of IO NP on differentiation of stem cells. The aim of the present study was to investigate the possible toxic role of different doses of IO NP in di...

We report the stimulation of neurogenesis and myogenesis of human mesenchymal stem cells (hMSCs) on the surfaces of biodegradable hydrogels with different stiffness. The hydrogels were composed of gelatin-hydroxyphenylpropionic acid (Gtn-HPA) conjugate were formed using the oxidative coupling of phenol moieties catalyzed by hydrogen peroxide (H(2)O(2)) and horseradish peroxidase (HRP). The stor...
متن کاملNanotopography-induced changes in focal adhesions, cytoskeletal organization, and mechanical properties of human mesenchymal stem cells.
The growth of stem cells can be modulated by physical factors such as extracellular matrix nanotopography. We hypothesize that nanotopography modulates cell behavior by changing the integrin clustering and focal adhesion (FA) assembly, leading to changes in cytoskeletal organization and cell mechanical properties. Human mesenchymal stem cells (hMSCs) cultured on 350 nm gratings of tissue-cultur...

We report the establishment of a novel platform to induce myogenic differentiation of human mesenchymal stem cells (hMSCs) via focal adhesion (FA) modulation, giving insights into the role of FA on stem cell differentiation. Micropatterning of collagen type I on a polyacrylamide gel with a stiffness of 10.2 kPa efficiently modulated elongated FA. This elongated FA profile preferentially recruit...
متن کاملFocal Adhesion Kinase (FAK) Involvement in Human Endometrial Remodeling During the Menstrual Cycle
Background: Endometrial remodeling occurs during each menstrual cycle in women. Reports have shown that, in a variety of cell types, processes such as proliferation, signaling complex formation and extra cellular matrix remodeling require a cytoplasmic tyrosine kinase, focal adhesion kinase (FAK). The present study has focused on the expression pattern of FAK in human endometrium during the men...
